"DIY" RESEARCH  ASSAYS

We are pleased to report a continuing strong growth in interest in our "do it yourself" (DIY) research assay protocols for HIV-1-p24gag and HIV-1 gp120 env. The antibody pairings and related substrates for available protocols are shown in the table below :

Capture Sheep Antibody

Enzyme Conjugate of          HIV-1-p24 gag MAb and HIV-1 gp120 MAb

Substrate Approx Working Range of DIY Assay

PROTOCOL 1 Anti-HIV-1-p24 gag
(D7320)

Alkaline Phosphatase Conjugate
(BC1071-AP)
Oxoid's "AMPAK" enzyme cycling system involving NADPH, diaphorase and alcohol dehydrogenase (Aalto Code K6200) 0.3 to 10ng/ml
PROTOCOL 2 Anti-HIV-1-p24 gag
(D7320)

Alkaline Phosphatase Conjugate
(BC1071-AP)

Chemiluminescent substrate enhancer from Tropix (cat No. EL100CX) 0.01 to > 40ng/ml
PROTOCOL 3 Anti-HIV-1p24 gag
(D7320)
Biotinylated Conjugate
(BC1071-BIOT)
p-Nitrophenyl phosphate (Sigma Cat. No. 104 -105) 0.1 to 3ng/ml

PROTOCOL 4 Anti-HIV-1-gp120env
(D7324)

Any specific gp120 MAb can be used together with an enzyme labelled anti-antibody - our DIY assay uses Alkaline Phosphatase as label Oxoid's "AMPAK" enzyme cycling system involving NADPH, diaphorase and alcohol dehydrogenase (Aalto Code K6200) 1 to 100ng/ml

The above listed DIY asays are being used widely by AIDS research laboratories in Europe, the United States and Africa for monitoring HIV-1 replication in a range of research applications.   These include monitoring the effect of drug inhibitors of HIV-1 replication, including inhibitors of virus entry and reverse transcription and measuring the effect of virus neutralising antibodies.   In short, one of our DIY assays can be used any time that an endpoint is needed for the extent of HIV-1 replication in cell culture.   The particular DIY assay selected will depend on the sensitivity needed.

Our AIDS research customers are using our DIY protocols because they are technically adequate and yet considerably cheaper than using higher sensitivity commercial assays that are licensed for clinical use. Our reagents and DIY protocols are intended only for research use and not for clinical use.                                                    BACK TO HIV PRODUCTS
